Tumor tissues that express any specific form of IGFs more than adjacent wild-type cells are potential candidates for an antisense gene therapy against the expression of that specific form of IGF. Paraffin sections containing these cancer cells and adjacent wild-type cells were examined by immunohistochemical technique using mouse IgG-1 antihuman IGF-I, or antihuman IGF-II as primary antibodies. Normal liver tissue, known as the largest producer of both forms of IGFs, was used as a positive control. The staining pattern of both IGF-I and IGF-II in the liver was highest in the cytoplasm of normal hepatocytes. The majority of the tumors except for papillary thyroid carcinoma and nasopharyngeal carcinoma strongly expressed IGF-I. Only a few samples of the tissues studied expressed a low level of IGF-II, if any. This raises a further working proposal that the blockade of IGF-I synthesis in these tumors may help to delay the tumor progression so that an effective immune response can be established and destroy the tumor.

Software                     Cholangiocarcinoma cells were isolated from biliary fluid aspirated from the lumen of bile duct upstream from the intrahepatic obstruction in subjects inflicted with cholangiocarcinoma using endoscopic retrograde cholecystopancreatomy (ERCP). These established cultured cells were confirmed for their epithelial origin by their intensely staining with primary antibody against cytokeratin. By using flow cytometric analysis, these cultured cell were positively stained for insulin-like growth factor 1 (IGF-1) and Fas (CD95) using the respective specific monoclonal antibodies. The findings of IGF-1 expression suggested that IGF-1 could mediate an autocrine action in the proliferation of these cells. The downregulation of IGF-1 within these cells may have a therapeutic potential in the respective subjects. The observation of Fas on these cells suggested that these cancer cells could enter into apoptotic stage in responsive to contact with Fas ligand (CD95L).

Data Recovery                     The expression of Fas ligand (FasL) on the surface of tumor cells has been proposed as a mechanism in which tumor cells evade cell-mediated immune responses. The direct interaction between FasL and Fas, the specific receptor on the surface of target cells, will activate a set of endogenous mediators and eventually lead to apoptosis within target cells. Upon direct contact with FasL on tumor cells, T cells that harbor Fas on their cell surface can undergo apoptosis. Hence the population of tumor-specific T cells within the proximity of tumor cells was controlled by such mechanism of contact inhibition. We proposed that the suppression of FasL expression or the overexpression of Fas in tumor cells could improve the autogeneic immune response against the tumor cells. This hypothesis warrants the construction of separate expression of systems for the antisense RNA of FasL and for the mRNA of Fas. We have synthesized two novel episomes, pAnti-FasL and pFas that can express the antisense FasL mRNA and the Fas mRNA within eukaryotic cells. Both episomes were modeled after pAnti-IGF-1, the episome that expresses the antisense RNA of IGF-1 upon the activation with ZnSO4. The pAnti-IGF-1 has been used in gene therapy against cancer. The cDNA construct of either FasL or Fas obtained from Dr. Shigekazu Nagata was subcloned to replaced the IGF-1 sequence in antisense and sense orientation respectively within the pAnti-IGF-1. Both novel episomes were successfully constructed. The expression of the antisense RNA of FasL from pAnti-FasL and the expression of Fas mRNA were controlled by the addition of ZnSO4. The function of both pAnti-FasL and pFas in transfected cultured tumor cells were confirmed using flow cytometry with the respective specific monoclonal antibodies. These episomes are crucial in the study for the dominance of tumor-derived immunosuppression based on contact inhibition. Such in vitro evidence of improved anti-cancer immune response could warrant the use of both episomes for gene therapy. Both episomes contain all components necessary for clinical application since they are derived from pAnti-IGF-1, the episome already approved for such purpose.
